Tail Suspension Test SOP:
1. Pre-Test Preparation: Ensure that all equipment is operational before the experiment, including the video camera, suspension apparatus, data acquisition system, and software. Keep ambient lighting consistent throughout the session. Transport animals to the testing room in their home cages and allow at least 30 minutes of acclimation in a quiet environment to minimize stress-induced variability.
2. Apparatus Cleaning: Before each trial, wipe down the suspension hook, panel, and surrounding area with 75% ethanol to eliminate any residual odors or biological traces. Allow surfaces to air dry fully before introducing the next subject.
3. Camera and Setup Configuration: Mount the camera facing the testing panel, ensuring the entire body of the mouse will be visible during suspension. Connect the camera to the recording system using a video capture card. Adjust camera focus and position to avoid blind spots or motion artifacts in the video feed.
4. Software Setup: If behavior is recorded and analyzed using software, create a new experiment and define the testing zone within the camera’s view. Calibrate the video feed to match the physical dimensions of the apparatus. Set the trial duration to 6 minutes, and define behavioral parameters as required for analysis.
5. Behavioral Testing Procedure: Suspend the mouse by the tail using adhesive tape, securing it approximately 1–2 cm from the tip. Affix the tape to the suspension hook so that the mouse is freely hanging and cannot touch any surfaces or climb its tail. Begin video recording immediately after suspension. Observe the animal for 6 minutes, during which periods of immobility and struggling will be recorded manually or automatically, depending on the setup.
6. Post-Session Cleaning and Data Management: After the session, gently remove the tape and return the animal to its home cage. Clean the suspension area and hook again with 75% ethanol. Save and organize all video files and tracking data. Ensure proper labeling of experimental groups for subsequent analysis.
