1. Software Setup and Arena Calibration: Connect the camera to the computer using a video capture device and launch EthoVision XT. Create a new experiment and select a data storage path. In the video settings, adjust the camera angle and focus to ensure the entire elevated plus maze is clearly visible. Define the central platform and four arms (two open, two closed) as distinct zones within the arena settings. Configure detection thresholds and object size to ensure accurate tracking of the animal’s movement. It is recommended to run a short pilot trial with one rat to confirm proper tracking and make any necessary adjustments to lighting or detection parameters.
2. Starting the Test:Place the rat gently in the center of the maze, facing one of the closed arms. Start video recording and set the trial duration to 5 minutes.
3. Behavioral Tracking: EthoVision will automatically track and record the time spent in each arm and the number of entries into open and closed arms. If tracking manually, use a stopwatch to record open-arm time and tally entries.
4. Ending the Session: After 5 minutes, stop the recording and save the behavioral data. Export the metrics of interest (e.g., time in open arms, number of entries).
5. Return the rat to its home cage.
6. Cleaning: Thoroughly clean the maze using quatricide before testing the next animal to remove scent cues and ensure consistency.
