1. Pre-Test Preparation
Ensure that all equipment, including the camera, acquisition system, and software, is functioning correctly before the session. Turn on the lights and maintain consistent ambient lighting throughout the experiment to reduce variability. Animals should be transported to the testing room in their home cages and allowed to acclimate for at least 30 minutes before testing.
2. Arena Cleaning
Before each session, thoroughly clean all open field arenas using 75% ethanol to eliminate any residual odors or biological traces from previous animals. Wipe down the walls and floor of each chamber with ethanol-soaked wipes, and allow the surfaces to air dry completely.
3. Camera and Mounting Setup
Connect the camera to the tracking system using a video capture card. Adjust the camera angle and position to make sure all arenas are clearly visible in the software’s video feed without distortion or occlusion. Confirm that the camera is level and properly focused.
4. EthoVision XT Software Setup
Open the EthoVision XT software and create a new experiment configuration. Define each of the four open field arenas as separate zones, each measuring 40 cm × 40 cm. Within each arena, define a center zone of 20 cm × 20 cm, centered geometrically. Use the live camera feed to verify that the defined zones match the physical layout accurately. Set the session duration (e.g., 5–10 minutes) and confirm detection settings.
5. Behavioral Testing Procedure
Place each animal gently into the center of its respective arena, minimizing handling and avoiding external disturbance. Avoid any sudden noise or vibrations during the test.
6. Post-Session Cleaning and Data Management
After the session ends, stop the recording. Remove the animals and return them to their home cages. Immediately clean the arenas again using 75% ethanol, removing any feces, urine, or scent marks.
