SOP:
1. Pre-Test Preparation: Ensure that all equipment, including the video camera, data acquisition system, and behavioral tracking software, is functioning properly. Turn on the room lights and maintain consistent ambient lighting to minimize variability. Animals should be transferred to the testing room in their home cages and allowed to acclimate for at least 30 minutes before testing.
2. Maze Cleaning: Before each trial, thoroughly clean the entire T-maze—including the start arm, choice arms, and walls—using 75% ethanol to remove any residual odors or biological traces from previous animals. Allow all surfaces to air dry completely before introducing a new animal.
3. Camera and Mounting Setup: Connect the camera to the tracking system via a video capture card. Adjust the camera height and angle to ensure that the full T-maze is clearly visible in the video feed. Ensure that the maze is centered and not distorted in the software interface, and that the camera is focused and level.
4. EthoVision XT Software Setup: Launch the EthoVision XT software and create a new project or load an existing T-maze configuration. Define the start arm and both goal arms as individual zones. Verify zone positions using the live camera feed to ensure alignment with the physical maze. Configure detection settings for accurate animal tracking, and set the trial duration (commonly 5–10 minutes per animal).
5. Behavioral Testing Procedure: Gently place the animal at the start arm of the T-maze, facing the intersection. Avoid sudden movements or noises. Allow the animal to explore and make a choice between the two arms. Depending on the specific protocol (e.g., spontaneous alternation, reward-based task), follow appropriate trial logic and timing.
6. Post-Session Cleaning and Data Management: After each trial, stop the recording and return the animal to its home cage. Immediately clean the T-maze again with 75% ethanol to remove any waste or odor cues. Save and back up the tracking data, and prepare the maze for the next subject.
